FACSCalibur Flow Cytometer

Description

FACS CaliburThe BD Biosciences FACSCalibur is a 2 laser flow cytometer that can be used to simultaneously analyze up to 4 fluorescent parameters. The instrument is designed to support a wide range of applications with high resolution and sensitivity of both fluorescence and scatter. The fixed excitation optics includes a 15 mW 488 nm argon laser and a 635 nm red diode laser. Emission optics are FL1 (FITC) 530/30, FL2 (PE/PI) 585/ 42, FL3 (PerCP) 670LP, FL4 (APC) 661/16.

Instrument Use

The instrument is available for sample acquisition on a sign-up basis. Fees are listed below. Prior to use, individuals must meet with Sarah or a designated member of the lab, read the Flow User's Guide, and be trained in basic cytometry operation. Flow users will be trained in basic cytometer useincluding start-up, shut-down and cleaning, calibration (FACSComp), PMTadjustments, compensation, instrument and cell settings, establishing a template, data acquisition, data analysis using CellQuest Pro Software, and use of training modules. It is up to the User to practice independently after initial training. If additional training is needed, Users must contact Sarah to schedule more hands-on training at the training rate. It should be noted that there is not a dedicated flow technician in this laboratory. Before being allowed to schedule use of the instrument, individuals will need to demonstrate the ability to acquire samples independently.

Data Analysis

There is a Mac computer setup for data analysis using CellQuestProSoftware in Goss 110. Please save your data files and templates to a USB drive and analyze your data at this separate station. Sign-up for the data analysis station is on a first-come, first-serve basis (please do not sign up for this station using the online scheduler).   • Sign-up in online at:

  • You will need to login. If you have any problems please call the Cytometry Core (2-9716) or lab (7-8025).
  • Please ONLY schedule the amount of time you need. (ie: DO NOT block out 2 hours for 10 tubes.)
  • DO NOT RESERVE ONLINE FOR ANALYSIS TIME. Analysis is available at any time in Goss 108 on a first-come, first-serve basis.
  • Acquisition has priority over analysis and individuals doing analysis on the instrument computer will be bumped to allow sample acquistion by other users. Therefore, make every attempt to do analysis on the designated computer.
  • Your time may be bumped a bit if the laboratory gets a CLINIC sample.
  • You must give 24 hours notice if you need to cancel your reservation. Your lab will be assessed a late cancellation fee of $25 and your PI will be contacted if you are a no-show. Repeat offenders will have privileges revoked.

Billing

  • Billing will be calculated by the hours of use rounded to the nearest half hour.
  • We use a 100-W form. You must check out a 100-W from your departmental representative, have your PI or designated lab manager sign-off and put the up-to-date ACCOUNT information on the form.
  • You may check one 100-W form out per month and log ALL of your USAGE on that form. Billing will be submitted on a monthly basis.
